Question : Describe the steps of PCR technique

Answer»

Solution :PCR `rarr` Polymerase chain reaction includes the three steps and they are :
(a) Heat denaturation : Heating of DNA at about `91^(@)C` The heating breaks the hydrogen bonds to MAKE single strand (SS) DNA. The DNA molecule with more G-C pairs, need higher temperature.
(b) ANNEALING : Pairing of primers to the single strand (SS) DNA segment. It requires temperature at about `55^(@)C`.
(c ) POLYMERISATION : Temperature is RAISED to `72^(@)C`. The Tag polymerase adds dNTP's behind the primer on the single strand (SS) DNA.
