Answer» Solution :a. The isolated PLASMID MUST have the following features. i. Origin of replication ii. Selectable market iii.Cloning sites b. Recombination can be identified using antibiotic resistance. When INSERTION is done at tetracyclineresistance GENE site, the transfor-mants will not show antibiotic resistance when placed in tetracycline containing MEDIUM . Recombination can also be identified using insertional inactivation method. The plasmid will give a blue colour to the colony if there is no inserted foreign DNA . |
