Significance of .heat shock. method in bacterial transformation is to FACILITATE uptake of DNA through transient pores in the bacterial cell wall. In heat shock method the bacterial CELLS are placed with the recombinant DNA on ice followed by PUTTING them at 42 °C and again on ice. This cycle is repeated resulting in formation of transient pores in bacterial cell wall through which the recombinant DNA ENTERS the cell. The pores then get closed.
