Question : One of the methods by which recombinant DNA can be introduced into host cell is by 'heat shock' treatment (electro-poration). Write down other three important methods by which we can introduce alien DNA into host cells . |
Answer» Solution :i. Micro-injection : Recombinant DNA is DIRECTLY injected into the nucleusof animal cell. ii. Biolistics or gene gun : Cells are bombarded with high velocity microparticles of gold or tungsten coated with DNA. iii.By disarmedpathogen vectors :These vectors are allowed to INFACT the cell, which TRANSFERS the recombinant DNA into the host . |

Question : One of the main objective of biotechnology is to minim.ise the use of insecticides on cultivated crops. Explain with the help of a suitable example how insect resistant crops have been developed using techniques of biotechnology. |
Answer» Solution : By using the techniques of biotechnology, Insect RESISTANT crops have been developed. Some strain of Bacillus thuringiensis (Bt) produce protein crystals which contain a toxic insecticidal protein that kill some insects like lepidopterans, coleopterans and DIPTERANS, etc. Specific Bt toxin genes were isolated from Bacillus thuringiensis and CLONED in bacteria. Specific_ Bt toxin genes obatained from Bacillus thuringiensis are used in several crop plants like cotton by providing them resistance from insects. Bt toxins arinsect-group specific, coded by a GENE called cry which are of various types. PROTEINS encoded by the genes cry IAc and cry IIAb control the cotton boll worms, that of cryIAb controls corn borer. |

Question : One of the applications of biotechnology is the production of insect resistant crop plants, Justify the statement with referance to Bt. Cotton. |
Answer» Solution :Crops are made which are more tolerant to environmnental stresses LIKE cold, heat drought, salt etc. BT. Cotton. a. Bacillus thuringensis produce crystal proteins called cry proteins which are toxic to larvae of insects like tobacco, budworm, army worm, bettles and mosquitoes. b. Cry protein exist as inactive prototoxin and gets converted into active toxin when ingested by the insects, as the alkaline pH of the gut solubilises the crystals. c. The activated toxin binds to the SURFACE of epithelial CELLS of midgut and creates pores. d. This causes swelling and LYSIS of cells leading to the death of the insects. e. The gene (cry genes) encoding this protein are isolated from the bacterium and incorporated into several crop plants like cotton, tomato, corn, rice, soyabean etc. f. The proteins encoded by the following cry genes control the pest given against them. i. crylAC and cryllAb it controls cotton bollworms. ii. crylAb controls corn borer. |
